Q. 2 Which of the following is required in the formation of the structure of Haemoglobin?
 A

Copper

 B

Tyrosine

 C

Histidine

 D

Leucine

Ans.
C
Explanation:

The Beta-chain of Hb begins with valine and histidine. Both these amino acids are important in the synthesis and structure of heamoglobin.

Alignment according to helical designation makes homology evident:

Residue F8 is the proximal heme-linked histidine, and the histidine on the distal side of the heme is E7.

The iron atom is linked by a coordinate bond to the imidazole nitrogen (N) of histidine F8.

The E7 distal histidine, on the other side of the heme plane, is not bonded to the iron atom but is very close to the ligand-binding site.

Q. 3

Along with succinyl CoA which of the following amino acid serve as starting material in heme synthesis?

 A

Lysine

 B

Leucine

 C

Glycine

 D

Alanine

Ans.
C
Explanation:
The two starting materials of heme synthesis pathway are succinyl CoA and glycine. Pyridoxal phosphate is needed to activate glycine. ALA synthase catalyzes the conversion of succinyl CoA and glycine to delta aminolevulinate. This is the rate limiting enzyme  in porphyrin biosynthesis in the mammalian liver. Synthesis of ALA occurs in mitochondria.

Last 3 steps of heme synthesis occur in the cytoplasm. The 3 enzymes which catalyse the reactions in cytoplasm are ferrochelatase, protoporphyrinogen oxidase and coproporphyrinogen oxidase.
  • Coproporphyrinogen oxidase catalyze conversion of coproporphyrinogen III to protoporphyrinogen III.
  • Protoporphyrinogen oxidase catalyze conversion of protoporphyrinogen III to protoporphyrin III.
  • Ferrochelatase catalyze conversion of  protoporphyrin III to heme by incorporating heme into protoporphyrin.

Q. 4

All of the following enzymes involved in heme synthesis are found in the mitochondria, EXCEPT:

 A

Ferrochelatase

 B

Protoporphyrinogen oxidase

 C

Coproporphyrinogen oxidase

 D

Uroporphyrinogen decarboxylase

Ans.
D
Explanation:
Uroporphyrinogen decarboxylase which catalyze the conversion of Uroporphyrinogen III to Coproporphyrinogen III is located in the cytoplasm. All the other 3 enzymes ferrochelatase, protoporphyrinogen oxidase and coproporphyrinogen oxidase are found in the cytoplasm.

Q. 5

Which of the following is required in the initial stage of the synthesis of haemoglobin?

Ans.

Explanation:
